Abstract

OBJECTIVE To investigate the difference in expression of aromatase and cyclooxygenase-2 in human eutopic endometrial cells derived from endometriosis and hysteromyoma and to explore the effect of medicated serum of SLW on aromatase and cyclooxygenase-2.METHODS After the model of eutopic primary cultured endometrial cells of endometiosis and hysteromyoma in vitro was successfully established,the changes of aromatase and cyclooxygenase-2 mRNA and protein were measured by RT-PCR and western blot respectively before and after treatment of medicated serum of SLW,and the level of secretion of E2 and prostaglandin E2 was also measured by ECLIA and RIA.RESULTS The expression of aromatase and cyclooxygenase mRNA,protein,secretion of estradiol and prostaglandin E2 in Hysteromyoma endometrium were weaker,as compared with that of eutopic primary cultured endometrial cells of endometiosis,which were taken as control group (P0.05). After the treatment of medicated serum of 5.0,2.5 g·kg-1·d-1 of SLW,the expression of aromatase and cyclooxygenase-2 mRNA and protein in eutopic endometrial cells of endometriosis were found significantly lower than of the control group(P0.01). Meanwhile,the expression of aromatase and cyclooxygenase-2 protein and aromatase mRNA in 1.25 g·kg-1·d-1 medicated serum group decreased,as compared with that of the control group(P0.01). However,The low dose group had no inhibitory effect on the expression of Cyclooxygenase-2 mRNA (P0.05).Compared with that of the control group,the level of the secretion of estradiol and prostaglandin E2 in cultural supernanant derived from endometrial cells of endometriosis in all three different dose of SLW groups was obviously lower(P0.05).CONCLUSION The medicated serum of SLW could inhibit the expression of aromatase and cyclooxygenase-2 in human eutopic endometrial cells derived from endometriosis and decrease the concentration of estradiol and prostaglandin E2 in local endometrium.
