Abstract
The 8 th Biennial Meeting of ASGO 2023 Even with diverse treatment, most individuals with advancedstage OC recurrence.In previous studies, immune escape is critical to tumorigenesis.Studies have shown that tumor-infiltrating lymphocytes may associate with clinical outcomes and drug resistance in OC. T cells are a tiny group of primarily tissue-resident lymphocytes that possess both innate and adaptive features.These distinguishing characteristics make them particularly appealing candidates for the development of novel cellular therapies aimed at tumor development.Thus, we investigate the interaction between extrinsic T cells and OC cells.Methods: In our study, clear cell adenocarcinoma, a poorly differentiated serous adenocarcinomas and endometrioid/ clear cell histological subtype were investigated in vitro and in vivo.To determine the effect of T cells in vivo, T cells were delivered intraperitoneally or intravenously into advanced severe immunodeficiency mice under an intraperitoneal tumor growth model.We also testified the possibility of T cells combined with the clinical chemotherapeutic drugs for further clinical application.And the mice were treated with T cells, and chemotherapeutic drugs.Six to 8 weeks following tumor injection, all animals were euthanasia and necropsied.Following the sacrifice of the mice with intraperitoneal tumor growth model, we extracted various intraabdominal organ tissues and examined the distribution of metastatic tumors.The survival rate of tumor bearing mice were statistic with the log-rank test.Results: Our results have shown that T cells effectively eliminate 3 different types of OC cell lines in vitro.The in vivo result revealed T cells suppress the tumor growth and enhance the effect of chemotherapy. T cells injection significantly inhibited the growth rate of 3 OC cell tumors in vivo.The T cells intraperitoneal treatment group had significantly lower metastasis than intravenous treatment.The survival rate of T cells intraperitoneal treatment group combing with low dose chemotherapy was not inferior to fulldose chemotherapy growth.The combing treatment group had significantly lower light signal than the other groups.Conclusion: T cells-immunotherapy can enhance the elimination of different types of OC tumor cells and improve survival in vivo. T cells has indicated its possibility of being an usher in a new era of cell therapy.
