Abstract

Estrogen exerts its effects through binding to its receptor and with the assistance of cofactors such as GREB1. Polymorphisms in the gene encoding this protein (GREB1) are potential biomarkers of susceptibility and prognosis for endometriosis, as alterations in GREB1 morphology, function, and synthesis resulting from these variants may lead to changes in estrogen signaling. The objective of this study was to investigate whether the estrogen-responsive GREB1 gene polymorphism (rs13394619 A>G) influences susceptibility to endometriosis, as well as its disease phenotype and associated symptoms, in Brazilian women. A retrospective case-control study of 442 women (221 cases and 221 controls) was conducted at referral hospitals in Rio de Janeiro. The GREB1 rs13394619 A>G polymorphism was genotyped using a validated TaqMan assay by real-time PCR. In the present study, we found that the GREB1 rs13394619 variant was associated with an increased risk of endometriosis, particularly with the deep infiltrating form of the disease, and with the presence of cyclic intestinal complaints in Brazilian women.
