Thompson Seedless was studied by foliar application of four treatments used: T0 (Control), T1 (Biostimulant elicitors based formulation), T2 (Calcium salt), and T3 (combination of Biostimulant and Calcium based formulation) at three spray schedules of (2 days before harvest (DBH), 10 DBH, and 10+2 DBH). The results demonstrated that Treatment T1 showed a significant decrease in berry rotting compared to the control group in both the 2 DBH and 10 DBH groups. The browning of the rachis was significantly reduced by T1 treatment, resulting in maintenance of rachis colour until 8 DAH. The application of T2 at 10 DBH led to a significant drop in berry shattering. In the 2 DBH group, T2 exhibited the highest reductions in berry cracking. In addition, T1 consistently exhibited elevated levels of Total Soluble Sugar showing a significant increase in the 2 DBH group. T1 demonstrated superior efficacy in managing weight loss when applied to plants at 2 DBH. Hence, these findings highlight the capability of Velabs's Biostimulant and calcium-based formulation to effectively address postharvest issues in table grapes via a synergistic approach. India, with its profound cultural legacy and strong connection to viticulture, is among the leading nations in grape production globally. The cultivation of grapes spans over an expansive area of 171 thousand hectares, resulting in an output of 3781 thousand metric tonnes 1 . Nevertheless, even if the nation has a strong output, the issue of postharvest losses continues to be a significant worry. This problem is mostly caused by the decay of berries or bunches, which is assisted by several diseases throughout the processes of handling, long distance transportation and storage 2 . The postharvest susceptibility of grapes and other fruits to rotting is a complex problem, impacted by several variables such as climatic conditions, handling and storage procedures 3 . Comprehending the complexities of grape postharvest physiology and the microbial dynamics linked to decay is essential for developing efficient mitigation measures. Table grapes, although being non climacteric fruit with a relatively low rate of physiological activity 3 have significant postharvest losses during storage and long distance transit 4 . During postharvest handling, there are several quality issues that can occur, including rapid moisture loss, which leads to the drying and browning of the rachis 5, 6 , weight loss 7 , berry shattering, wilting and shriveling 8 . These problems can result in both quantitative and qualitative losses. The major cause of the loss of grape’s natural defence and their increased vulnerability to rot and degradation is believed to be due to improper handling practices 9 . The decay of berries or clusters in grapes is mostly caused by fungi, including Botrytis cinerea , Aspergillus spp., and Penicillium spp 10–12 . These pathogens exploit fruit that has been compromised or weakened, often during harvesting or transit, to proliferate and induce decay. The widespread presence of these pathogens highlights the urgent need to investigate innovative methods to improve the shelf life and reduce the amount of spoilage. Plant signalling molecules and defensive response activation elicitors are now being considered as a means to enhance the resistance of grapes to postharvest deterioration. This represents a shift in thinking that has occurred recently 13 which may be due to the restricted use of postharvest fungicides in fruits due to health and environmental concerns. By stimulating the inherent defence mechanisms of grapevines, these tactics provide a proactive method to enhance the fruit's ability to withstand stress and prolong its shelf life. Compounds such as salicylic acid 14 , jasmonic acid 15 , and L-Phenylalanine 16 have been shown to efficiently induce acquired resistance and promote the activation of defense-related genes. As a result, there is an elevated resistance to attack from pathogens 17 . Moreover, the incorporation of defence response activation elicitors such as chitosan 18 , methyl jasmonate 19 and oligosaccharides 20 offers a potential approach to improve postharvest quality. These elicitors may activate the production of substances that shows negative response against microorganisms, strengthen the structure of the cell wall, and regulate the functioning of enzymes that control oxidative stress 21 . These approaches have great promise for reducing postharvest losses and guaranteeing the supply of high quality grapes to customers. Additionally it is important to consider the application time, frequency, concentrations of the preharvest elicitors 22 based on the abiotic factors and cultivars. Hence the objective of this study was to assess preharvest treatments that use defence response activation elicitors critically for their ability to reduce postharvest rotting and its associated losses in grapes. Materials and Methods The investigation was conducted in a vineyard located in Ambevani village, Nashik district (20.245457, 73.898775) during the March 2024 season. The four year old table Grapes cv. Thompson seedless plants grafted on Dogridge ( Vitis champinii L.) were selected for experiments. The experiments includes three main treatment i.e., foliar spray scheduled on 2 days before harvest (DBH), 10 DBH and 10 + 2 DBH with the following sub treatments of T0 - Control, T1 - Biostimulant based formulation, T2 - Calcium salt, T3 - combination of Biostimulant and Calcium based formulation. Each vines were sprayed with approximately half liter of the spray fluid. This novel Biostimulant and calcium based formulation was developed with organic acid, amino acid and calcium salt by Velabs (Vegrow, Bengaluru, India) for grapes. The bunches were harvested according to spray schedules in perforated liner bags and then transported to Vegrow’s Pack house at Sakore, Nashik ( 20.143380, 73.942673) and stored in perforated crates under room temperature. The observation were recorded at 2 days interval from 0 days after harvest (DAH) till 8 DAH. The number of rotten berries per bunch were counted and weighed and expressed as percentage of rotting. The rachis browning were evaluated on a visual score from 1 to 4, i.e., 1 - fresh and green rachis, 2 - browning initiation, 3 - significant browning and 4 - severe browning 23 . The berry shattering was calculated by weight of the loose berries to its total bunch weight and expressed in percentage. Cracked berries in individual bunches were weighed and expressed as percentage of cracked berries per bunch. Total soluble sugar (TSS) was estimated as brix level by digital refractometer (Parisa technologies, Mumbai, India). The physiological loss in weight (PLW) was calculated based on initial weight basis and expressed in percentage. The experiment was setup in Factorial CRD with three replications and the analysis of variance (ANOVA) was performed using OPSTAT 24 and the graphs were developed in GraphPad Prism version 10.0.0 for Windows, GraphPad Software, Boston, Massachusetts USA. Results Berry rotting The percentage of berry rotting increased gradually with increase in storage days in all spray groups and in all treatments. The highest % of rotting was seen in control (T0) in all cases. All the treatments were effective in controlling berry rotting while the least rotting was observed in T1 and T3 in spray schedule of 2 DBH and 10 DBH respectively. Remarkably, in the plants with spray schedule of 10 + 2 DBH, both treatments T1 and T3 exhibited similarly low levels of rotting. In overall, the data shows that treatment T1 led to a substantial decrease in rotting in both the 2 DBH and 10 DBH groups, as compared to their respective controls (Fig. 1 ) (Table. 1). Rachis browning The rachis browning accelerated with increase in days after harvest among all treatments and in all spray groups. In the spray schedules of 2 DBH, 10 DBH, and 10 + 2 DBH, the rachis achieved a score of 4, which indicates full browning, at the end of the 8 DAH. Both T1 and T3 in the 2 DBH group maintained a completely green rachis until 4 DAH, with T1 showing a lower score than the control by 8 DAH. T1 and T3 underwent a transition from a completely green state to the early phases of browning in the 10 DBH group. At 8 DAH, T1 had the lowest score when compared to the control group. Within the 10 + 2 DBH group, there was a progressive increase in browning. At 8 DAH, both treatment T1 and T3 achieved a score of 3.5, which was comparable to the control group. Based on the total mean data, treatments T1 in the 2 DBH group were effective in maintaining the pigmentation of the rachis until 8 DAH (Fig. 2 ) (Table. 2). Berry Shattering Among all spray schedules (2 DBH, 10 DBH, and 10 + 2 DBH), T0 consistently had a highest level of berry shattering, whereas T2 consistently had the lowest rate of shattering in all three groups. Remarkably, the lowest percentage of shattering was detected in T2, which was sprayed at 10 DBH. This indicates a decrease of 67.16% in berry shattering as compared to the control groups that correspond to them (Fig. 3 ) (Table. 3). Berry cracking T0 constantly showed a greater rate of berry cracking throughout all spray regimens. In contrast, the prevalence of cracking was lowest in T2 among the 10 DBH group and both T1 and T2 within the 2 DBH group. T2 and T3 showed similar trends in reducing the cracking in 10 + 2 group. The overall pooled data represented shows that in the 2 DBH group, both T1 and T2 exhibited a significant decrease in cracking. Similarly, T2 showed reductions in the 10 DBH and 10 + 2 DBH as compared to their respective controls (Fig. 4 ) (Table. 4). Total Soluble Sugars The TSS content showed minor variations as affected by the preharvest treatments during the storage period in all spray schedules. The control (T0) had the lowest TSS content, whereas T1 consistently showed greater sugar levels in all three spray scheduled plants. Specifically, in the spray given 2 DBH, there was a notable 20.90% rise in brix in T1 as compared to the control. These findings indicate that treatment T1 significantly improved the sugar buildup in the berries in plant sprayed 2 DBH than on plants sprayed 10 and 10 + 2 DBH (Fig. 5 ) (Table. 5). Physiological loss in weight There was a significant loss in weight throughout the storage period in all samples. The control (T0) had the highest PLW across the three spray groups. Specifically, among the plants sprayed with 10 DBH, T2 shown enhanced effectiveness in lowering weight loss when compared to the control while T1 maintained lesser weight loss in 2 DBH and 10 + 2 DBH sprayed groups. Nevertheless, the most effective method to control weight loss was shown to be T1 when sprayed on plants 2 DBH (Fig. 6 ) (Table. 6). Discussion Fresh produce including grapes have poor self life and are prone to postharvest losses due to fungal decay, and also due to other physiological process like respiration rate, hormonal cross talks, storage conditions etc. The use of postharvest fungicides is an effective method of controlling the fungal decay 25 but due to their concerns of pathogen resistance, human health and environmental safety, the use of postharvest fungicides and synthetic chemicals has been restricted 26 . The preharvest treatment of bunches with Velabs’s biostimulant showed to reduce berry rotting especially when applied 2 DBH. This formulation was developed with the compositions of organic and amino acids that may act as a elicitors molecules that triggers natural defence system like phenylproponoid pathways and thereby reduces the grapes susceptibility to rotting. Similar results on reducing fungal decay was observed by preharvest application of L-phenylalanine in strawberry, mandarin and mangoes 27 . Results revealed the preharvest application of Chitosan, Fulvic acid and Salicylic acid combinations reduced berry rotting in grapes cv. Thompson Seedless 28 . The significant reduction in berry rotting was seen in the bunch dipping of 1% calcium propionate 3 DBH in grapes 29 . The first priority in terms of postharvest loss is to prevent berry decay, but notably, rachis browning is the next important storage parameter as consumer preference is also a cause of postharvest losses 30 . The preharvest application of T1 was found to maintain the rachis colour untill 8 DAH. This may be due to improved antioxidant activity that can mitigate osmotic stress, retard chlorophyll degradation and hence maintain the colour. Similarly, Salicylic acid and oxalic acid treatment maintained rachis colour in grapes 10 . Preharvest foliar treatment of salicylic acid reduced decay incidence and maintained rachis colour in grapes cv. Flame Seedless 31 . Foliar spray of Calcium salt (T2) particularly at 10 DBH showed lowest berry cracking and shattering. This may be due to the poor calcium absorption and mobility to berries after veraison due to nonfunctional xylem and lower transpiration rate of the fruits 32 . Cell wall swelling in berries due to pectin solubilization and loss of calcium in cell wall complex decreases the elasticity of berry skin and induces cracking 33 . Hence foliar spray of calcium salt (T2) may reduce berry cracking by increase the calcium pools especially in cell wall complexes and thereby maintain osmotic pressure, inhibit tissue swelling and improving berries skin mechanical properties. Similarly studies on calcium nitration application at verasion reduced berry cracking and shattering in grapes 34 . Table grapes are highly prone to berry shattering due to abscission during storage and transport resulting in significant loss in commercial value 35 . Application of prohexadione calcium (Pro-Ca) reduced the level of ABA by modulating endogenous hormonal levels and berry shattering in grapes 36 . Hence Calcium salt spray may be involved in ABA signalling and inhibit the rate abscission zone formation thus maintain the berry integrity within the clusters. Similar results were also reported by Zhu et al 37 as nano calcium and calcium chloride spray on 2 weeks before harvest maintained higher berry detachment force and lower abscission percentage in grapes cv. Thompson Seedless. The biostimulant based treatment (T2) showed to maintain higher TSS when applied 2 DBH. This may be possible by organic and amino acid based formulation which may regulate starch sugar conversion by upregulating the activity of α and β amylase. Our results were comparable with the earlier reports on increase in TSS content by amino acid application in apple 38 . Shafiee et al 39 reported that the physiological loss in weight of fruits is due to the combined effect of postharvest metabolic activities, respiration and transpiration. The results of our study showed that PLW in grapes was observed to increase with increase in storage period. The foliar spray of T2 on 2 DBH showed to reduce weight loss as compared to control and other spray schedules. This may be due to the elicitors which maintained better physiological conditions like reduced transpiration and respiration rate of grapes after harvest. Similar results on weight loss were reported in kiwi 40 and apple 41 . Conclusion The use of preharvest interventions, such as Velabs's biostimulant elicitor based formulation derived from organic and amino acids (T1) and the administration of calcium salt by foliar spray (T2), exhibits considerable promise in mitigating diverse postharvest issues encountered in table grapes. T1 demonstrated favourable results by decreasing berry decay, maintaining rachis colour, improving TSS and controlling weigh loss especially when applied at 2 DBH. Additionally, the application of T2 at 10 DBH was shown to be successful in minimizing berry cracking and shattering after harvest, which might influence the cell wall elasticity and abscission processes. Thus Velabs Calcium salt and biostimulant elicitor based formulation can be recommended as preharvest spray on 10 DBH and 2 DBH respectively to synergistically mitigate the above address postharvest issues in grapes. Furthermore, there is potential for further research to explore the precise mechanisms and gene regulation that trigger the activation of natural defence systems by these innovative formulation. Nevertheless, even if the nation has a strong output, the issue of postharvest losses continues to be a significant worry. This problem is mostly caused by the decay of berries or bunches, which is assisted by several diseases throughout the processes of handling, long distance transportation and storage\u003csup\u003e2\u003c/sup\u003e. The postharvest susceptibility of grapes and other fruits to rotting is a complex problem, impacted by several variables such as climatic conditions, handling and storage procedures\u003csup\u003e3\u003c/sup\u003e. Comprehending the complexities of grape postharvest physiology and the microbial dynamics linked to decay is essential for developing efficient mitigation measures. Thompson Seedless was studied by foliar application of four treatments used: T0 (Control), T1 (Biostimulant elicitors based formulation), T2 (Calcium salt), and T3 (combination of Biostimulant and Calcium based formulation) at three spray schedules of (2 days before harvest (DBH), 10 DBH, and 10+2 DBH). The results demonstrated that Treatment T1 showed a significant decrease in berry rotting compared to the control group in both the 2 DBH and 10 DBH groups. The browning of the rachis was significantly reduced by T1 treatment, resulting in maintenance of rachis colour until 8 DAH. The application of T2 at 10 DBH led to a significant drop in berry shattering. In the 2 DBH group, T2 exhibited the highest reductions in berry cracking. In addition, T1 consistently exhibited elevated levels of Total Soluble Sugar showing a significant increase in the 2 DBH group. T1 demonstrated superior efficacy in managing weight loss when applied to plants at 2 DBH.
