Poly (ADP-Ribose) polymerase inhibitors have efficacy in a fraction of platinum-resistant ovarian cancer patients

Abstract
BACKGROUD:Platinum-based chemotherapies are widely used as first line treatments for ovarian cancer after debulking surgery. Poly (ADP-ribose) polymerase inhibitors (PARPis) have also been approved for the treatment of some types of ovarian cancer, such as ovarian cancer with BRCA1 and BRCA2 mutations or deletions. Both platinum and PARPi treatments have been shown to relate to homologous recombination deficiency. However, whether PARPis are efficient in platinum-resistant patients is still unclear. METHODS:In this study, we enrolled 36 ovarian cancer patients who received treatments of both platinum and PARPis and analyzed the demographic, clinical, and pathological data. RESULTS:The results indicated that the patients who are sensitive to platinum had better responses to PARPis (P=0.004 ** , 99%CI: 0.002-0.005). PARPis show efficacy in a fraction of platinum resistant patients. Moreover, the earlier usage of PARPis was associated with a lower risk of death (P=0.006 ** , 95%CI: 0.66-0.630). CONCLUSIONS:Platinum-resistant patients were less responsive to PARPis. PARPis are also options for platinum-resistant patients.
