Study on rat model of premature ovarian failure induced by Tripterygium wilfordii polyglycoside

Abstract

Objective:To explore the feasibility of applying Tripterygium wilfordii polyglycoside(TWP) to establish rat model of premature ovarian failure(POF). Methods:Thirty Wistar female rats of SPF level were randomly divided into control group and model group.The rats in model group were treated with pouring TWP suspension liquid(50mg/kg daily) into stomach for 14 days;while the rats in control group were treated with pouring normal saline of the same volume into stomach.The changes of body weight,uterine wet weight,ovarian wet weight,oestrous cycle,serum levels of estradiol,follicle stimulating hormone(FSH),luteinizing hormone(LH),ovarian histomorphological changes,and uterine histomorphological changes in the two groups were observed. Results:The manifestations of oestrous cycle of rats in model group were different types of disorders;the body weight of rats in model group was higher than that in control group(P0.05);the serum level of estradiol in model group was significantly lower than that in control group(P0.05);the serum level of LH in model group was significantly higher than that in control group(P0.05);the serum level of FSH in model group was higher than that in control group,but there was no significant difference(P0.05);the thickness of endometrium and ovarian area in model group were significantly lower than those in control group(P0.05);the numbers of follicles and corpus luteum in unit ovarian area in model group decreased significantly(P0.05),the number of atretic follicles increased significantly(P0.05). Conclusion:It is feasible to establish rat model of POF with TWP.
