Application of chemical pretreatment and molecular identification of Actinomycetes isolates

Abstract

Actinomycetes are an interesting group of filamentous Gram-positive bacteria that includes a wide range of several habitats and produce compounds of pharmaceutical and industrial interest. Molecular identification based on 16S rRNA gene sequencing is a reference approach for studying their taxonomic diversity. This research work is about Actinomycetes isolates from soil samples after chemical pretreatment with sodium dodecyl sulfate and yeast extract designed to facilitate selective growth. Genomic DNA was extracted, and the 16S rRNA gene was amplified using four pairs of primers: the universal primers 243F/ A3Rb and two specific primers, Sm6F/Sm5Rb  and M2F/ A3Rb. The amplified fragments were sequenced by the Sanger method, analysed by SkyBLAST than a phylogenetic trees, was constructed to elucidate the evolutionary relationships among the strain studies. Tha analysis revealed that the obtained sequences high identity with several Streptomyces species including Streptomyces argenteolus, Streptomyces rosa and Streptomyces olivaceus. The study demonstrates that the use of chemical pretreatment and 16S rRNA molecular identification proved to be an effective approach for the precise characterisation of Streptomyces sp, which became the basis for various innovations, especially in the field of medicine.
