Effect of CO_2 pneumoperitoneum on protein expression of MMP-2 and TIMP-1 of endometriosis lesion in the rat

Abstract
Objective To evaluate the effect of CO2 different pneumoperitoneum pressure on protein expression matrix metalloproteinase-2(MMP-2)and tissue inhibitor of metalloproteinase-1(TIMP-1)of endometriosis lesion.Methods The SD rats were divided into randomly three groups:control group,10 mmHg and 20 mmHg CO2 pneumoperitoneum after endometriosis model being founded successfully.The pressure continued for about 1h.The MMP-2 and TIMP-1 in EMs lesion were detemined at 1w,2w and 6w.Results Compared with the control group,the protein expression level of MMP-2 in the two CO2 pneumoperitoneum groups were decreased significantly in post-operation one week(P0.001).And the effect could be continued until two weeks after pneumoperitoneum intervention.Moreover the expression level of MMP-2 in 10mmHg group was lower than that in 20 mmHg group pneumoperitoneum(P0.001).The protein expression of TIMP-1 increased in CO2 groups,with the higher expression in 10mmHg group(P0.001).The expression level of MMP-2 was increased and TIMP-1 was decreased gradually with time after CO2 pneumoperitoneum intervention,and there were no difference compared with that of the control group six weeks post-operation(P0.05).Conclusion The expression of MMP-2 is decreased and TIMP-1 is increased in the early period CO2 pneumoperitoneum intervention.The low CO2 pneumoperitoneum pressure is superior to the high pressure group in inhibition of aggression of EMs lesion.
