The goal was to assess the effects of growth regulators gibberellic acid (GA3) and indole-3-acetic acid (IAA) on seed germination and early seedling development in vitro and in vivo.The seeds were hydro-primed and surface-sterilized for 48 hours before being treated with different concentrations of GA3 (5 and 10 ppm) and IAA (5 and 10 ppm). The seeds were hydro-primed and surface-sterilized for 48 hours before being subjected to various concentrations of GA3 (5 and 10 ppm) and IAA (5 and 10 ppm). Embryo rescue was performed using Murashige and Skoog (MS) media for in vitro germination, with in vivo germination observed for comparison. The results showed that the maximum germination rate (77.78%) and better shoot and root growth were attained under in vitro conditions with GA3 at 5 ppm, with early shoot emergence at 8.40 days and shoot length reaching 6.63 mm. In vivo germination rates, however, were much lower, with just 6.67% germination in seeds treated with GA3 at 5 and 10 ppm and 0% in those treated with IAA. GA3 at 5 ppm was determined to be the best concentration for increasing germination and development of Calcutta 4 x Ney Poovan tetraploid hybrid seeds, especially in vitro.
