Notably, substances such as methylmercury, dioxins, polychlorinated biphenyls, and more recently per- and polyfluoroalkyl substances (PFAS) have garnered attention in this context (EFSA 2005, 2020). In the case of farmed fish, the careful consideration of the origin and source of feed materials is crucial for maintaining low contamination levels (Malisch, 2017 ). On the other hand, for wild fish and seafood, factors such as the quality of the environment—including sediments and water columns— the benthic, demersal and the pelagic zones coupled to the trophic level of the biota of alimentary interest play a pivotal role in determining the bioaccumulation of priority pollutants like PFAS (SCHEER, 2022). Recent findings from the US-FDA PFAS monitoring program in food (Young et al., 2022 ) substantiate other observations from convenient sampling activities from the Mediterranean Sea (Hlouskova et al., 2013 ; Barhoumi et al., 2022 ), indicating that benthic biota, such as clams and crabs, are more susceptible to contamination with medium- to long-chain PFAS compared to other seafood resources. Moreover, in the context of the revised Water Framework Directive, the European Commission Scientific Committee on Health, Environment, and Emerging Risk (SCHEER, 2022) proposed a Relative Potency Factors grid for 24 PFAS encompassing also C10-C18 perfluoro carboxylic acids. This grid aims to facilitate a comprehensive cumulative PFAS health risk assessment for individuals consuming fish and seafood from fresh and coastal waters and would represents an update of that proposed by EFSA in its 2020 PFAS opinion (EFSA, 2020) because includes additional congeners beyond the former four PFAS —PFOS, PFOA, PFHxS, and PFNA— considered by EFSA, keeping immuno-toxicity as shared toxicological end-point. This paper seeks to discuss the implications of applying such a grid to PFAS intake assessments within the framework of One Health considerations and the establishment of safe and sustainable food systems, aligning with the 17 Sustainable Development Goals outlined by the United Nations. Materials and Methods PFAS Occurrence Data in fish and seafood. Based on literature (Dellatte et al., 2014 ) and the Italian Food Consumption Database 2018 available on the EFSA Zenodo platform (EFSA, 2022), we identified the following benthic and demersal seafood species as representative of the most consumed ones at the national Mediterranean Sea (FAO 37 Area) level: clams, mollusks like squid and cuttlefish, and crustaceans (deep water rose-shrimp). This set was complemented with farmed sea bream. Occurrence data were obtained from the results of the EU FP7 “Perfood” project (Hlouskova et al., 2013 ). For clams, data were extracted from sample 1.1 of the US-FDA PFAS 2022 monitoring plan (Young et al., 2022 ), representing the average PFAS contamination recorded in such commodity. Fish and seafood consumption Figures for fish and seafood consumption were extracted from the Italian food consumption database 2018 (EFSA, 2022), specifically for people aged 10–74 years (mean and P95 g/kg bw per day). The recovered values for the considered categories were as follows: squid, cuttlefish, octopus (0.76; 2.87); clams (0.35; 0.88); crustaceans (0.54; 1.37); farmed sea bass and sea breams (1.48; 3.11). Relative Potency Factor-based PFAS intake scenarios Toxicological-based intakes were computed according to EFSA Relative Potency Factor (RPF) for 4 PFAS (EFSA, 2020), and to Scientific Committee on Health, Environmental and Emerging Risks of the European Commission (SCHEER) RPF for 24 PFAS (SCHEER, 2022) respectively. As reported in the SCHEER opinion, the same provisional Health Based Guidance Value (HBGV) of Σ PFAS 4.4 ng /kg body weight per week (equivalent to Σ PFAS 630 pg /kg body weight per day), tailored on immuno-toxicity was assumed. In the proposed scenario, a lower-bound approach was chosen, posing no quantified results equal to 0. Hazard indices were computed as the ratio between the estimated intake from each of the fish and seafood categories considered and the PFAS HBGV. Results Table 2 shows the output of the RPF based intake scenario for EFSA and SCHEER for the considered fish and seafood categories, while Fig. 1 illustrates the corresponding hazard index with respect to mean and P95 consumption figures in Consumers Only from the Italian 2018 database. We selected results from analytical methods capable of matching performances suitable for the panel of PFAS considered within the same sample (EURL POP, 2022). The limit of quantification, particularly aiming for values around or below < 10 ng/kg fresh weight, was another crucial criterion, especially concerning PFAS with assigned higher RPFs (refer to Table 1 ). We chose data from the recent US-FDA 2022 survey on clams (Young et al., 2022 ) because this food category is underrepresented in the literature from the Mediterranean area, in terms of number of samples and PFAS congeners analysed. This holds true under both SCHEER and EFSA toxicological grids, specifically designed for sensitive and vulnerable groups, without accounting for inputs from food categories other than fish and seafood (such as milk, eggs, drinking water, vegetables). Table 2 reveals that the ratios of computational outputs from the SCHEER and EFSA RPF grids indicate SCHEER PFOA equivalent derived values for 24 PFAS are x19, x1.8, x7.4, and x2.7 higher in cuttlefish, clams, crustaceans, and farmed seabass, respectively compared to the EFSA values set on 4 PFAS, only. This is due to the varying presence of ≥ C9 perfluoroalkyl carboxylates (PFCAs) in the selected categories (Table 1 ). Consequently, applying the SCHEER grid would significantly increase the Hazard Index compared to that of EFSA, particularly for those commodities with a quantifiable presence of congeners with higher RPFs (see Fig. 1). In perspective, what represented above would highilight the following challenges: Sampling activity and database representativness challenge. Sampling activity should be more risk oriented, possibly accounting for the origin and provenience of fish and seafood commodities, with a more detailed appropriate georeferencing with respect to the current FAO areas. In doing this, adequate number of samples to draw PFAS contamination baseline values, as well on time updates of fish and seafood consumption should be achieved, to orient the risk. Analytical challenge . Under the SCHEER RPF grid, is rather evident that analytical methods should be tailored also accounting for the highest RPFs, to prevent unquantified results could generate a stronger underestimation of the exposure and of the related risk. Currently, the European Commission Reccomendation 2022/1431 indicates as analytical performance criteria LOQs set below or at 300 pg/g for PFOS, PFOA, PFNA and PFHxS in crustaceans and molluscs and at 100 pg/g in fish meat, to support intake assessment only for the four PFAS considered by EFSA. However, Member States should also consider testing for the presence in food of long chain PFCAs and of emerging PFASs, such as FOSA, the acid form of F53B, Capstone, Fluorotelomer sulfonates, thus broadening the spectrum of PFAS to be monitored to overlap the SCHEER list. For PFNA, for instance, according to the SCHEER RPF = 10 (Table 1 ), LOQs should be lowered at least of a factor of 10 with respect to that for PFOA. In detail, if SCHEER opinion indicates a cumulative PFAS 24 EQS biota food hh of 77 pg/g fresh weight, LOQs should be oriented in the range 0.1–1.0 pg/g, accounting for PFAS occurrence and the related RPF. Economic and social challenge . The considerations above suggest higher sampling and analytical costs, involving time-consuming analyses (processing larger test portions to reach target LOQs), and potentially requiring the use of high performing but expensive instruments with low detection limits. While such costs may be manageable for official bodies tasked with PFAS analysis, the sampling strategy may not cover all fish and seafood species and provenances. Nevertheless, food business operators (FBO) internal checks could focus on ensuring compliance/incompliance assessments with regulatory limits. It's worth noting that in the European Union the regulatory limits for fish and seafood are 2–3 times higher than the LOQs currently recommended by the European Commission for intake assessments above reported. As example, for Mediterranean Sea fish and seafood species, we record a cumulative Σ PFOS,PFOA, PFHxS PFNA Maximum Limit of 8 ng/g in seabass meat, 5 ng/g in clams and crustaceans, up to 45 ng/g in anchovies; no specified limits have been set for squid and cuttlefish species (EU Regulation, 2022) . Internal checks performed by FBO are likely focused on the four regulated PFAS, around the maximum level: the validation of methods testing for compliance/incompliance is typically conducted in the range from ½x to 2x of the Maximum Level. Additional indirect costs would involve the accreditation of the method for analytes other those currently regulated. This could significantly hinder the generation of representative and reusable data from FBO internal check activities, particularly for local, small, short-chained food production and distribution systems that may struggle to afford the analytical costs. The last but not least impact is on the so called “blue growth”, where freshwater and coastal areas are considered a privileged environment for the production of food according to the 17 United Nations Sustainable Developmental Goals, as matter of the reduced water and carbon footprints. Under the prospected scenarios, we could envisage limitation in local fish and seafood consumption, discussed in more details under the following One Health challenges paragraph. One Health Challenges: Accountability of risk managers in preventing intolerable exposures with attention paid to vulnerable groups: The discrepancy between regulatory level of PFAS contamination in the order of ng/g and those requested to protect vulnerable groups (1–2 order of magnitude lower) could hamper the accountability of environment and food&feed risk managers, especially if a clear, timed and effective strategy to reduce PFAS environmental pollution of water as factor for a sound fish and seafood production is not exploited. Regulatory maximum limits in food should to be set to ensure a high level of human health protection, but also account for a risk/benefit balance between food security (prevent food and nutritional losses due to not compliant food put out of the market) and food safety. In some cases, when the food resources recognise short supply chains, or subsistence economy environments, the release of advisories for local populations is recommended, along with a georeferenced indication of fish and seafood species more prone to be heavily contaminated. At present, apart hot-spot areas, recommendation for PFAS in this sense to the general population have not been released yet, in contrast with those already made for Dioxins and polychlorinated biphenyls in some countries (e.g. Swedish Food Agency 2023 ). The other need is a tighter coordination between environment and food monitoring activities on geo-referenced basis in monitoring plan, with sampling activities adequate to characterise at the same time the Environmental Quality of the waters where molluscs and crustacean farming and/or harvesting take places, and biota of alimentary interest PFAS burden. This could be combined with a community science approach. Conclusion The high exceedance of the Hazard Index for PFAS intake from fish and seafood highlights the need for a robust reduction and substitution policies of PFAS production and use. This is crucial to avert unsustainable consequences for food safety and security, preventing the loss of accountability for official bodies entrusted with ensuring environmental quality and animal and human health. This is in response to the sustainability mismatch between risk management and risk assessment. Currently, the European Commission Reccomendation 2022/1431 indicates as analytical performance criteria LOQs set below or at 300 pg/g for PFOS, PFOA, PFNA and PFHxS in crustaceans and molluscs and at 100 pg/g in fish meat, to support intake assessment only for the four PFAS considered by EFSA. However, Member States should also consider testing for the presence in food of long chain PFCAs and of emerging PFASs, such as FOSA, the acid form of F53B, Capstone, Fluorotelomer sulfonates, thus broadening the spectrum of PFAS to be monitored to overlap the SCHEER list. For PFNA, for instance, according to the SCHEER RPF\u0026thinsp;=\u0026thinsp;10 (Table\u0026nbsp;\u003cspan refid=\"Tab2\" class=\"InternalRef\"\u003e1\u003c/span\u003e), LOQs should be lowered at least of a factor of 10 with respect to that for PFOA. The expansion of the analytical per- and poly-fluoroalkyl substances (PFAS) panel with a toxicological characterisation necessitates an update of PFAS intake assessment and management within the context of cumulative exposure. Benthic and demersal seafood such as clams, squid, and cuttlefish, are more prone to contamination with C9-C14 perfluoro-carboxylic acids, exhibiting the highest Relative Potency Factors in terms of immuno-toxicity, according to the grid recently proposed by the European Commission Scientific Committee on Health, Environment, and Emerging Risk. Based on the Italian food consumption database referred to demersal and benthic biota such as squid, cuttlefish, crustaceans, and clams, a toxicological-based intake scenario has been drafted to highlight the relevant contribution of such wild species to PFAS intake, well above the provisional health-based guidance value of 4.4 ng/kg body weight per week proposed by the European Food Safety Authority. Ensuring consistency between PFAS reduction and substitution policies, environmental quality standards for water bodies and the issuance of advisories and regulations regarding fish and seafood safe\u0026amp;sustainable intake is crucial.
