Additionally, it assesses their antioxidant and cytotoxic potential to explore their industrial and therapeutic applications. Methods The growth conditions of Streptomyces isolates were evaluated by varying temperature, pH, and incubation period. Temperature preferences were tested from 30°C to 60°C, while pH was adjusted between 5.0 and 7.5. Growth dynamics were monitored, with peaks identified between 48 and 120 hours. Antioxidant activity was measured using the DPPH assay, assessing radical scavenging potential. Cytotoxicity was evaluated using the brine shrimp lethality assay, and LC50 values were determined to gauge the potency of cytotoxic effects. Results Distinct temperature preferences were observed among the isolates, with most showing peak growth between 30°C and 40°C, typical for mesophilic organisms. Isolate DM 1 uniquely thrived at 60°C, suggesting high-temperature stability. pH optimization revealed that most isolates preferred slightly acidic to neutral conditions (pH 5.0 to 7.5). Growth peaks varied from 48 to 120 hours. Antioxidant activity was high in isolates YM 3 and DMP 3, also noted in GA 2 and CSV 2. Cytotoxicity assays showed that isolates CSV 1 and DMP 3 had the highest cytotoxicity, with lower concentrations yielding significant effects. Isolates GA 1, GA 2, and YM 4 also demonstrated potent cytotoxicity at lower concentrations over extended exposure, in contrast to lower cytotoxicity observed in CW isolates. Conclusion These findings provide insights into optimizing cultivation conditions and exploring potential applications in biotechnology and pharmaceuticals. These microorganisms are predominantly found in soil, where they play a critical role in decomposing organic matter and contributing to nutrient cycling (Viega et al ., 2004). The bioactivity of Streptomyces is heavily influenced by environmental conditions, which dictate their growth and metabolic processes. Understanding these conditions is crucial for optimizing their cultivation and harnessing their full potential for biotechnological applications. In Anyigba, Kogi State, the rich and diverse soil microbiota presents a promising source of novel Streptomyces isolates with potentially unique metabolic profiles. This region's soil environment, characterized by its specific thermal and pH conditions, offers a distinctive backdrop for studying the growth dynamics and bioactivity of these isolates. The present study aims to investigate the thermal optimization, pH sensitivity, and growth dynamics of Streptomyces isolates derived from soil samples in Anyigba. By systematically analyzing how these environmental factors affect the growth and bioactivity of the isolates, we seek to identify optimal conditions for maximizing metabolite production (Kaur et al., 2012 ). Additionally, this study will explore the bioactivity of these isolates, particularly their antioxidant and cytotoxic potential, which can inform their application in pharmaceuticals and other industrial processes. Through a comprehensive examination of temperature and pH preferences, growth patterns, and bioactive properties, this research contributes to a deeper understanding of Streptomyces ecology and its practical implications. The findings have the potential to advance the development of targeted cultivation strategies, enhance the yield of valuable metabolites, and uncover new bioactive compounds with significant therapeutic potential. Materials and Methods Isolation and Cultivation of Streptomyces Isolates Soil samples were collected from various locations in Anyigba, Kogi State, and processed for the isolation of Streptomyces species using standard techniques. Approximately 10 g of soil was mixed with 90 mL of sterile distilled water and serially diluted. The dilutions were spread on ISP2 agar plates (International Streptomyces Project) supplemented with cycloheximide to suppress fungal growth. Plates were incubated at 28°C for 7–14 days to allow colony formation. Isolates were then subcultured on fresh ISP2 agar for purification. Temperature Sensitivity and Growth Patterns For thermal optimization, purified Streptomyces isolates were grown on ISP2 agar plates and incubated at various temperatures ranging from 20°C to 60°C. Growth was monitored daily for 7 days. The optimal growth temperature for each isolate was determined by observing the temperature at which the isolate exhibited the highest growth rate. These findings are consistent with previous studies on the thermal adaptation of actinobacteria (Gao et al., 2010 ); (Schafer et al ., 2012). pH Optimization for Growth To determine pH sensitivity, isolates were grown in ISP2 broth adjusted to different pH levels (2.5, 5.0, 7.5, and 9.5) using hydrochloric acid or sodium hydroxide. The cultures were incubated at the optimal temperature for each isolate and monitored for growth over 7 days. Growth was assessed by measuring optical density at 600 nm. The preferred pH range for each isolate was identified based on maximum growth, in line with previous findings on Streptomyces pH preferences (Kieser and Hopwood, 2000 ); (Jermann et al., 2013 ). Growth Dynamics and Incubation Period Growth kinetics were assessed by incubating isolates in ISP2 broth at their optimal temperature and pH. Samples were taken at 24-hour intervals for up to 120 hours. Growth curves were generated by plotting optical density at 600 nm against time. The peak growth phase was determined for each isolate, reflecting variations in growth dynamics (Viega et al ., 2004). Antioxidant Activity Antioxidant activity was evaluated using the DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging assay. Isolates were cultured in ISP2 broth, and the supernatant was collected after 48 hours of incubation. The supernatant was mixed with a DPPH solution, and the decrease in absorbance at 517 nm was measured. The percentage of radical scavenging activity was calculated. Results were compared with known antioxidants as described by (Shahidi and Ambigaipalan, 2015 ) and (Blois, 1958 ). Cytotoxicity and LC50 Analysis Cytotoxicity was assessed using the brine shrimp lethality assay. Artemia salina nauplii were exposed to varying concentrations of metabolite extracts from Streptomyces isolates. After 24 and 48 hours, the number of surviving nauplii was recorded. LC50 values, representing the concentration at which 50% of the nauplii were lethally affected, were calculated and compared with potassium dichromate as a reference cytotoxic agent. GA 1: Exhibits a granular, dry appearance with convex elevation and off-white-greenish aerial pigmentation. This aligns with the commonly observed dry and granular texture of Streptomyces in natural soil environments (Bérdy, 2005 ). GA 2: Features a dry, smooth texture and convex elevation, contrasting with the irregular edge and straight spore arrangement, highlighting a less common variation in appearance (Goodfellow et al ., 2012). YM 1 and YM 4: Show granular and smooth textures with convex to concave elevations and cream pigmentation. This variation in elevation and pigmentation is consistent with findings by Kuster and Williams ( 1964 ), who noted that environmental factors can significantly influence morphological traits. CSV 1 and CSV 2: Present smooth and dry textures with convex elevations and cream pigmentation, indicating uniformity within this farm's isolates, similar to observations by Loria et al. ( 2006 ). DM 1 and DM 3: Show varied appearances, with differences in pigmentation (grey, white) and edge types (fuzzy, regular), which might be indicative of the diverse and harsh conditions of refuse dumps affecting Streptomyces morphology (Hirsch et al., 2004 ). CW 1 and CW 2: Displayed a rough texture with varying pigmentation and spore arrangement. The rough texture is less common and might be a response to the specific soil conditions in the cashew farm environment (Kumar et al., 2014 ). Sugar Utilization (Table 2 ) The sugar utilization patterns highlight the metabolic diversity among the isolates. GA 1 and GA 9: Utilized a broad range of sugars, similar to other studies showing the metabolic versatility of Streptomyces strains from diverse environments (Hopwood et al., 2011 ). YM 1 and YM 2: Demonstrated varied sugar utilization, with YM 1 showing extensive sugar utilization compared to YM 2, reflecting the potential for strain-specific metabolic capabilities (Dye et al., 2007 ). CSV 1 and CSV 3 utilized multiple sugars, consistent with findings that Streptomyces strains from similar environments exhibit high metabolic adaptability (Chater, 2006 ). DM 3 utilized a wide range of sugars, indicating robust metabolic pathways potentially adapted to the refuse dump's nutrient variability (Kieser et al., 2000 ). CW 1 and CW 2 utilized a wide array of sugars, reflecting metabolic adaptability, which is crucial for survival in variable environments (Tabor and Tabor, 1984 ). Biochemical Tests (Table 3 ) The biochemical tests reveal varying physiological traits among the isolates. All isolates were gram-positive, consistent with the Gram-positive nature of Streptomyces (Kutzner, 1981 ). GA 2 and DM 1 showed negative results, suggesting strain-specific differences in metabolic capabilities (Garrity et al., 2005 ). Most isolates tested positive for catalase, indicating their ability to break down hydrogen peroxide, a common trait among Streptomyces (Madigan et al., 2009 ). Oxidase results varied, which is consistent with the physiological diversity reported in Streptomyces (Bérdy, 2005 ). The Variability in coagulase and indole production among isolates highlights differences in enzymatic activities, which can impact their ecological roles and interactions (Goodfellow et al ., 2012). The findings align with previous research that demonstrates the diverse morphological and biochemical characteristics of Streptomyces isolates from various environments. For instance, the morphological variations are consistent with studies by Williams et al. ( 1983 ) and the metabolic versatility observed is similar to findings by Loria et al. ( 2006 ). Thermal and pH Optimization for Streptomyces Isolates Temperature Sensitivity and Growth Patterns: Streptomyces isolates show distinct temperature preferences, with most exhibiting optimal growth between 30°C and 40°C, which is consistent with their mesophilic nature (Gao et al., 2010 ; Schaefer et al ., 2012). This range aligns with previous findings on actinobacteria. However, isolate DM 1, with an optimal growth temperature of 60°C, is an outlier, indicating thermophilic properties that are atypical for Streptomyces. This unique trait suggests potential applications in high-temperature industrial processes, which has been less explored in prior research (Schaefer et al ., 2012). Further study into the mechanisms underlying this thermotolerance could enhance our understanding and application of thermophilic Streptomyces strains. Cultivation Techniques: To maximize metabolite production, Streptomyces isolates should ideally be cultivated at temperatures between 30°C and 40°C. Isolates demonstrating thermotolerance, such as GA 2 and DM 1, offer promising avenues for research into their thermal adaptation mechanisms. Such investigations could lead to the development of more efficient thermophilic strains for industrial applications (Waksman, 1938 ), potentially improving biotechnological processes by leveraging these temperature-adapted strains. pH Sensitivity and Adaptation: The optimal growth pH for Streptomyces isolates ranges from 5.0 to 7.5, supporting their mesophilic characteristics (Kieser et al., 2000 ; Jermann et al., 2013 ). Extreme pH levels (2.5 and 9.5) significantly hinder growth, emphasizing the need for pH control in cultivation. These findings corroborate previous studies highlighting the importance of maintaining a neutral to slightly acidic pH for optimal Streptomyces growth and metabolite production. Cultivation Strategies: For effective large-scale cultivation, it is crucial to implement pH buffering systems to sustain the optimal growth environment. This practice ensures consistent growth and maximum metabolite production, reflecting the need for precise pH regulation in biotechnological processes (Waksman, 1938 ). Growth Dynamics and Incubation Period: Growth patterns varied among Streptomyces isolates over time. Isolates like GA 1, GA 2, and GA 9 followed a typical growth curve with a peak followed by stabilization or decline, while YM isolates showed delayed peak growth phases. This variability highlights the necessity of optimizing incubation times for each isolate to enhance productivity and efficiency (Viega et al ., 2004). Bioactivity of Streptomyces Isolates: Antioxidant Activity: Assessment of antioxidant activity using the DPPH assay revealed high activity in isolates YM 3 and DMP 3. These isolates demonstrated significant antioxidant potential through mechanisms such as hydrogen atom transfer and scavenging of reactive oxygen species (Shahidi & Ambigaipalan, 2015 ; Blois, 1958 ). This positions Streptomyces isolates as valuable sources of natural antioxidants with potential health benefits. Cytotoxicity and LC50 Analysis: The brine shrimp lethality assay showed that isolates GA 1, GA 9, and YM 3 exhibited high cytotoxicity, with CSV 1 and DMP 3 demonstrating the most potent effects. These results are consistent with other studies on Streptomyces strains known for their bioactive compounds (Miao et al., 2019 ). Conversely, CW 1 and CW 2 showed lower cytotoxicity, indicating potentially less potent or fewer bioactive compounds (Katz & Baltz, 2016 ). LC50 values, indicating concentration-dependent cytotoxicity, suggest high cytotoxic potential, though generally, Streptomyces isolates displayed lower cytotoxicity compared to potassium dichromate (Jin et al., 2014 ). These findings align with the broad spectrum of bioactivity reported for Streptomyces secondary metabolites and support their potential as sources of novel anticancer agents (Skrzypek et al., 2018 ; Kaur et al., 2012 ; Strohl, 1996 ). Conclusion The study of Streptomyces isolates reveals significant insights into their thermal and pH preferences, growth dynamics, and bioactivities. Optimal conditions for growth and metabolite production involve precise temperature and pH control. The antioxidant and cytotoxic properties of Streptomyces metabolites highlight their potential for biotechnological and pharmaceutical applications. Continued research into these aspects will be crucial for harnessing the full potential of Streptomyces in various industrial and medicinal contexts.
