Causal Relationship Between Socioeconomic Inequality and Hormone-Related Gynecologic Diseases: A Two-Sample, Two-Step Mendelian Randomization Study
This study used two-sample, two-step Mendelian randomization with European GWAS summary statistics to test whether socioeconomic status exposures (education, income, and occupation) have causal effects on five hormone-related gynecologic diseases and to explore potential mediating pathways. Genetically predicted higher educational attainment—but not income or occupation—was associated with lower risk of endometriosis overall and specifically uterine endometriosis in both univariable and multivariable models; mediation analysis suggested major depression might mediate the education–uterine endometriosis relationship with a reported mediation proportion of 12.22%. The authors’ main limitation is the reliance on European genetic instruments and the MR assumptions inherent to this approach, which may limit generalizability and causal interpretation.
