( 1998 ) and Cote et al. (1995),we have developed an improved protocol for extracting nuclei from yeast cells.This study investigates the efficiency of a cell nuclei extraction protocol using image-based quantification methods.Microscopic images were analyzed before and after the extraction process using ImageJ-equivalent computer vision techniques. The results show a significant reduction in nuclei count post-extraction,demonstrating the effectiveness of the applied method.This method provides a valuable tool for biologists requiring efficient nucleus extraction from yeast.
