Abstract

Endometriosis remains dominated by hormonal suppression and surgery despite mounting evidence that immune dysregulation + inflammatory amplification drive lesion persistence, pain, fibrosis, and recurrence. This OSF project builds an open, falsifier-driven, Minimal Viable Endometriosis Loop (MVEL): a modular causal chain (nodes + feedbacks) connecting lesion seeding/attachment → regional immune clearance failure (macrophage/NK dysfunction, tolerance/checkpoints) → cytokine/NF-κB amplification → oxidative stress (iron/ROS) → neuroangiogenesis/pain sensitization → fibrosis/adhesions/remodeling, with explicit interfaces, biomarkers, and falsifiers for each node
