Lineage tracing reveals atoh7 positive and negative retinal ganglion cell populations in the zebrafish retina
This study used lineage tracing in wild type and atoh7 mutant zebrafish to map the retinal cell fates arising from transient atoh7 expression, employing atoh7:iCre transgenic fish crossed with ubi:Switch for permanent labeling, and validating results with live imaging and immunohistochemistry across embryonic, larval, and adult stages plus the adult brain. It found that the labeling recapitulated endogenous atoh7 onset and that 79% of total retinal ganglion cells (RGCs) in wild type zebrafish derive from atoh7+ progenitors. In atoh7 mutants, the authors confirmed a complete loss of RGCs and observed increased proportions of atoh7+/Pax6+ amacrine cells and increased total Prox1+ bipolar cells, while also detecting atoh7+ cells outside the eye in other central nervous system regions.
