Renoprotect effect of fucoidan on adriamycin-induced glomerulosclerosis in rats

Abstract

Objective: To investigate the effects and mechanism of fucoidan on adriamycininduced glomerulosclerosis in rats.Methods: Thirty-two Sprague Dawley rats were randomly divided into sham operation,glomerulosclerosis model,fucoidan treatment,and valsartan groups.The 24-hour urinary protein excretion,renal function were measured after 8 weeks.Renal pathology and immunohistochemistry were assayed to measure the expression of fibronectin(FN),collagen Ⅳ(Col Ⅳ),transformor growth factor β_1(TGF-β_1).RT-PCR was used to examine the TGF-β_1 mRNA and plasminogen activator inhibitor-1(PAI-1) mRNA.Result: The proteinuria,BUN,and serum creatinine declined significantly(P0.01),and the expression of FN,Col Ⅳ,TGF-β_1,TGF-β_1 and PAI-1 decreased by fucoidan(P0.01).Conclusion: Fucoidan plays an important role on prevention and treatment for glomerulosclerosis.
