The experimental study of the effects of mifepristone on adenomyotic cells in vitro

Abstract
Objective: To observe the effects of mifepristone on the cultured adenomyotic cells. Method:With MTT assay, the growth state of primary cell culture from adenomyotic implants of 11 women with adenomyosis was studied. The cells of each case were divided into four groups, three treatment groups (1.25 mg/L, 2.5 mg/L, 5.0 mg/L mifepristone, respectively) and one control group, cultured for 52 hours before the OD value was determined. Results: The OD values of 1.25 mg/L, 2.5 mg/L, 5.0 mg/L mifepristone and control group were (0.219±0.044),(0.185±0.038), (0.152±0.033) and (0.262±0.068) respectively. The OD value of 5.0 mg/L group was significantly lower than that of 1.25 mg/L group (P0.01), and both 5.0 mg/L and 2.5 mg/L groups had significantly lower OD values than the control group (P0.01). The inhibition rate of cell growth in 2.5 mg/L group (IR=29.4%) was 1.79 times as much as that in 1.25 mg/L group (IR= 16.4%), while in 5.0 mg/L group (IR=42.0%)it was 1.43 times as much as that in 2.5 mg/L group. Conclusion: Mifepristone could suppress the growth of adenomyotic cells in vitro in a dose-dependent manner.
