Combined detection of HE4 and CA125 is more sensitive and effective for early ovarian cancer diagnosis than either biomarker alone.

Abstract

Objective To observe the value of early diagnosis and sequelae of the combined and separated detection of HE4 and CA125 in ovary cancer. To discuss the relationship between HE4 and the pathologic type of the ovary cancer, and tumor staging. Methods ELISA was used to detect the level of HE4 in the serum and the electrochemiluminescence immunoassay(ECLIA)was used to detect the level of CA125.40 clinical cases of ovary cancer,30 clinical cases of benign ovary tumor,30 clinical cases of endometriosis and 40 clinical cases of healthy women were collected in order to compare the difference between those groups. Results The serum level of HE4 and CA125 in the group of ovary cancer was markedly higher than in other groups(P 0.05).The difference of serum level of HE4 in the groups of ovary endometriosis cyst and healthy women did not have a statistical meaning, but the level of CA125 in the ovary endometriosis cyst was high,if compared to the healthy women the difference had the statistical meaning. In the group of the ovary cancer, the level of HE4 before the operation was related to the patient age,and was positively related to the pathologic grading and clinical staging. The whole patients who undergo the ovary cancer surgical staging, after the surgical operation the level of HE4 was markedly lower than the level of HE4 before the operation(P 0.05).In the healthy women, the population of reference for the benign diseases of the ovary, when the serum level of HE4 is 67pmol/L,it was the best limit point value. The ROC-AUC for diagnosing the ovary cancer was 0.969,the sensitivity was 92.5%,the specificity was 98%.The combined detection of HE4 and CA125 had the highest sensitivity. Conclusion HE4 is a sensitive biomarker for diagnosing the ovary cancer. Its sensitivity is higher than the sensitivity of CA125.The combined detection of HE4 and C125 is more effective in diagnosing the ovary cancer. The serum HE4 can be used as the sensitive serum biomarker for detecting the clinical sequelae and the status in the postoperative period of the ovary cancer.
