Effect of rIL-10 on HLA-G expression in endometrial cells of patients with adenomyosis

Abstract
Objective:To explore the change of expression of human leukocyte antigen-G(HLA-G) via recombinat interleukin-10(rIL-10) induction in stromal cells and glandular epithelium cells of patients with adenomyosis.Methods:Laboratory study using the human endometrium stromal cells and glandular epithelium cells obtained from 25 patients with adenomyosis undergoing hysterectomy.HLA-G protein expression in two cells via IL-10 induction with Flow cytometry assay,Western blotting and Immunofluorescence.Quantitative analysis of HLA-G protein expression via IL-10 induction,according to Student's t-test.Results:After the intervention of the rIL-10 for 48 h,a simple intervention group of rIL-10(intervention group B),the HLA-G expression of endometrial glandular epithelial cells was increased(P0.05)IL-10R antibody and rIL-10 joint intervention Group(intervention group A) and rIL-10 intervention group(not intervention group) have no significant change in the amount expression of HLA-G(P0.05).Compared with stromal cells without IL-10 induction,HLA-G protein expression had a statistically significant decrease in these with IL-10 induction(P0.05),but it is opposite for glandular epithelium cells.After IL-10 induction,the positive expression rate of HLA-G expression in two cells did not have obviously change(P0.05).Conclusion:rIL-10 stimulation increased the HLA-G expression of adenomyosis endometrial glandular epithelial cells in patients.rIL-10 stimulation in patients with adenomyosis expression of HLA-G may be the role of IL-10 Rα which can make adenomyosis patients with ectopic endometrium induce immune inhibition
