Abstract

Objective:To assay the levels of estrogen and angiogenesis related factors in adenomyosis mice,and to investigate their relationship and the role in the development adenomysis.Methods:Adenomyosis mouse model was established by transplanting isologous anterior pituitary glands(PGs)into the right uterine lumen on 8-week-age old ICR mouse.The angiogenesis indices(CD31,MMP-2,and VEGF)and the estrogen effect factors(P450,COX-2,and ERα)were detected by the immunohistochemisty method.Results:1As compared with normal control group,in the eutopic endometrium,the expression of CD31 decreased,VEGF and MMP-2increased significantly in adenomyosis mice;there was no significant change of P450,ERα,and COX-2after3 months of modeling,but after 6months,there was a significant up-regulation in P450 and COX-2.2 In the eutopic endometrium,there was no significant difference in CD31,VEGF,and MMP-2levels between 3-month and 6-month groups,while the P450,ERα,and COX-2levels was significant higher after 6months.In the ectopic endometrium,CD31 and VEGF had no significant difference between 3-month and 6-month groups,but MMP-2and COX-2levels were increased after six months.3At the same period of adenomyosis,the changes of VEGF and MMP-2were more significant in eutopic endometrium than in ectopic site,while the changes of P450 in ectopic site were more significant.Conclusion:Changes of indicators for angiogenesis and estrogen pathway were found in different time and different site of endometrium,which suggest that they may play a role in the pathogenesis of adenomyosis.
