Two-colour Imaging Fluorescence Correlation Spectroscopy (ImFCS) probes the influence of the juxta-membrane actin-cortex
The study extends Imaging Fluorescence Correlation Spectroscopy (ImFCS) using bright, photostable fluorophores and a membrane-anchored actin reporter to quantify how the juxta-membrane actin cortex shapes plasma membrane dynamics in live cells. To overcome the mismatch between the membrane’s nanometre thickness and the imaging axial resolution (~100 nm), the authors engineer an actin-binding construct and perform two-colour, co-registered diffusion mapping, comparing it pixel-by-pixel with a non-binding mutant. The actin-binding reporter shows diffusion coefficients spanning over two orders of magnitude, and the comparison yields an actin-occupancy map indicating that the actin cortex modulates transport over more than 80% of the cell surface.
