We hypothesized that arboviral infections during pregnancy impair placental histomorphometry and key transporter systems implicated with fetal brain development. Placental samples (n=54) were collected from singleton pregnancies affected by chikungunya (CHIKV; n=14), dengue (DENV; n=10), zika (ZIKV; n=15), or healthy controls (HC, n=15). Histomorphometry and immunohistochemistry were performed, with quantification of xenobiotic, nutrient, thyroid hormone transporters, and cell turnover markers. Data were analyzed using ANOVA (p<0.05) and principal component analysis (PCA). CHIKV was associated with reduced syncytiotrophoblast and syncytial knot (SK) areas compared with HC, and reduced SK area versus ZIKV. DENV showed decreased SK area versus HC and ZIKV, reduced connective tissue area compared with HC and CHIKV, and increased fetal blood vessel (FBV) area versus HC. ZIKV was linked to reduced syncytiotrophoblast and increased FBV area versus HC. All arboviral infections showed reduced immunoreactive (ir) expression of the protective transporters P-gp and BCRP. Nutrient transporter expression was altered across groups: CHIKV exhibited reduced ABCA1, SNAT2, and GLUT1; DENV exhibited reduced ABCA1 and SNAT2, whereas ZIKV exhibited reduced ABCA1, SNAT2 and GLUT1 with increased SNAT4 versus HC. MCT8 and Ki67 ir-expression were elevated in CHIKV and ZIKV compared with HC (all p<0.05). PCA distinguished infected from uninfected groups based on placental functional markers related to fetal brain development, but not on histomorphometry. Arboviral-infected placentae showed distinct structural changes and reduced nutrient, hormone, and xenobiotic transporter expression, potentially increasing fetal brain vulnerability and long-term health risks even when neonatal symptoms are minimal.
