Study on the changes of cell apoptosis in guinea pigs uterine fibroid embolization with danazol alginate microsphere

Objectives To study the changes of cell apoptosis in uterine fibroid embolization with danazol alginate microsphere for female guinea pigs as myoma model.Methods A total of 24 female guinea pigs as myoma model were induced by estrogen and progesterone for 16 weeks,and then all female guinea pigs were divided into 3 groups randomly:including a control group(n=8),danazol alginate microspheres group(DKMG group n=8) and alginate microspheres group(KMG group n=8).Simulate clinical UAE-operation process through open surgery with female guinea pigs except control group.Four weeks after surgery,apoptotic cells of myoma were assessed for all groups by TUNEL assay and the expressions of Bcl-2 for myoma cells were evaluated by IHC.Results Apoptosis rate of DKMG,KMG and control group was(44.6±11.9)%,(33.7±7.3)% and(22.8±9.7)% respectively.Compared to the control group apoptosis rate of DKMG,KMG was higher,and the difference was statistically significant(P〈0.05).There were also significant differences between DKMG and KMG group.H-scores of Bcl-2 for DKMG,KMG and control group was 2.7±0.7,2.1±0.3 and 1.3±0.6 respectively,and the H-scores difference between DKMG,KMG and control group was statistically significant(P〈0.05).Conclusions According to the effect in our test,the therapeutic mechanism of danazol alginate microsphere after UAE includes apoptotic cells increase with danazol releasing locally and with ischemia-reperfusion caused by microsphere degradation.
