The paper surveys how non-coding RNAs, focusing on microRNAs miR-99 and miR-17-5p, are involved in reproductive-system pathologies and evaluates their expression changes in blood. Using qReal-Time PCR, the authors measured miRNA expression in plasma from patients with endometrial carcinoma, uterine cervix cancer, and endometriosis, and report an association between the expression levels of these selected molecules and the investigated conditions. The main caveat explicitly stated in the work is that the findings are based on expression analysis of only two microRNAs rather than a broader non-coding RNA panel. This paper is centrally about endometriosis — it includes endometriosis in its plasma miRNA expression analysis and links miR-99 and miR-17-5p expression changes to endometriosis as part of its diagnostic-potential overview.

Abstract

SÚHRN Nekódujúce RNA predstavujú funkčné molekuly ribonukleovej kyseliny tvoriace rozsiahly segment transkriptómu, ktorý sa v rôznej miere uplatňuje vo fyziologických a patologických procesoch. Preukazujúc nielen diagnostický potenciál sú oprávnene tieto molekuly predmetom rozsiahlych výskumov. Predkladaná práca poskytuje prehľad zapojenia nekódujúcich RNA molekúl do patológií reprodukčného systému a prakticky sa opiera o analýzu zmeny expresie vybraných microRNA (miRNA, miR), konkrétne miR-99 a miR-17-5p, v plazme pacientiek s karcinómom endometria, cervixu uteru a endometriózou prostredníctvom metódy kvantitatívnej Real Time polymerázovej reťazovej reakcie (qReal-Time PCR). Dosiahnuté výsledky práce preukázali súvislosť medzi mierou expresie vybraných molekúl a uvedenými patológiami, čím naznačujú potenciálne možnosti využitia nekódujúcich RNA molekúl v diagnostickej praxi. ABSTRACT Non-coding RNAs are functional ribonucleic acid molecules forming a large segment of the transcriptome, which is involved in physiological and pathological processes. Demonstrating not only diagnostic potential, these molecules are justifiably the subject of research. The present work provides an overview of the involvement of non-coding RNA molecules in the pathologies of the reproductive system and is practically based on the analysis of the expression changes of selected miRNAs, namely miR-99 and miR-17-5p, in the plasma of patients with endometrial carcinoma, uterine cervix and endometriosis by qReal-Time PCR. The achieved results of the work demonstrated an association between the expression levels of the selected molecules and the aforementioned pathologies suggesting the potential use of non-coding RNA molecules in diagnostic practice.
