Effect of IL-8 on pathogenesis of endometriosis

OBJECTIVE To explore the level and effect of IL-8 on pathogenesis of endometriosis.METHODS The patients were divided in different group on base of R-AFS standard,and the levels of IL-8 in peritoneal fluid and IL-8 mRNA in endomembrane tissue were detected by ELISA,real-time-PCR,respectively.The levels of and the effect of IL-8,anti-IL-8 for hyperplasy or restrain of stroma cells were detected by MTT.RESULTS The levels of IL-8 in peritoneal fluid and endomembrane tissue were(23.37 ± 8.23)pg/ml,(12.32 ± 4.27)pg/ml,respectively(P﹤0.05).However,the expression of IL-8 mRNA in estrin phase of endomembrane was 1.301 1 ± 0.268 9(log cDNA/log GAPDH),1.123 9 ± 0.273 8(log cDNA/log GAPDH).It was similar in two groups(P﹥0.05).The dose-time dependence was expressed between the level of IL-8 and hyperplasy of stroma cells,and the best concentration was at site of 10ng/ml.The proliferative effect of IL-8 on stroma cell was inhibited by anti-IL-8.CONCLUSION IL-8,a kind of autocrine growth factor and positive feedback factor,can promote the growth of stroma cells and play a key role on pathogenesis of endometriosis.
