Sequential treatment with relugolix combination therapy in women with symptomatic uterine fibroids that were inadequately managed with 52 mg levonorgestrel intrauterine system: the first report
This first report evaluated the clinical impact and safety of sequential relugolix combination therapy (relugolix CT: relugolix 40 mg plus oestradiol 1 mg plus norethisterone acetate 0.5 mg daily) added to ongoing 52 mg levonorgestrel intrauterine system (LNG-IUS) in four women aged 45–52 with symptomatic uterine fibroids and persistent bleeding despite more than 6 months of LNG-IUS monotherapy; some also had coexisting adenomyosis. All participants reported complete resolution of bleeding and improved quality of life after starting relugolix CT, with no adverse events observed over a mean follow-up of 11.2 months (range 3–24), and satisfaction was 9.4/10. A key limitation is the very small sample size and lack of a comparator group, as this is a first report. Relevance to endometriosis/adenomyosis: the abstract notes that some enrolled women had coexisting adenomyosis, though the paper’s main focus is sequential non-surgical management of symptomatic uterine fibroids.
