Changes of contents of type I collagen and type III collagen in cardinal ligament of uterus of prostaglandin E_2-induced rabbit pelvic inflammation models

Abstract
Objective:To explore the effects of different administration times of prostaglandin E2 on the changes of contents of type Ⅰ collagen and type Ⅲ collagen in cardinal ligament of uterus of prostaglandin E2-induced rabbit pelvic inflammation models.Methods:The pelvic inflammation models were established by injecting prostaglandin E2(the same dose and the same concentration) into rabbits.At 2,4 and 8 weeks after establishment of pelvic inflammation models,6 rabbits were selected from each group randomly,then their cardinal ligaments of uterus were obtained.Staining and paraffin imbedding were used in the study;polarization microscope and electron microscope were used to observe the contents and changes of type Ⅰ collagen and type Ⅲ collagen.Results:Polarization microscope could observe the difference between type Ⅰ collagen and type Ⅲ collagen effectively.There was significant difference in the contents of type Ⅰ collagen and type Ⅲ collagen between normal pelvic ligament and inflammatory ligament(P0.01).It was confirmed that prostaglandin E2 could promote the secretions of collagens on the fourth week.There was no significant difference in the result between electron microscope and polarization microscope,the main manifestations included that symptoms of pelvic inflammation aggravated with administration time extension of prostaglandin E2,the synthesis of type Ⅰ collagen and type Ⅲ collagen at different time periods in cardinal ligament of uterus increased at first,and then decreased rapidly,showing a unimodal change.Conclusion:Prostaglandin E2 changes the differentiation and activity of fibroblast in cardinal ligament of uterus by regulating the metabolisms and proportions of type Ⅰ collagen and type Ⅲ collagen,which indicates that pelvic inflammation may participate in the occurrence of pelvic floor dysfunction.
