Experimental Approach The expression of THBS1 in macrophages was assessed using single-cell RNA sequencing and Western blot analysis. The therapeutic potential of THBS1 blockade in aortic perivalvular inflammation (AP) was evaluated through the application of TAX2, a THBS1/CD47-inhibiting peptide. A rheumatoid arthritis (RA) model was employed to validate the conserved regulatory role of THBS1 in inflammatory processes. Key Results In the context of acute pancreatitis (AP), macrophages exhibit a significant upregulation of THBS1. TAX2 enhances macrophage efferocytosis, thereby ameliorating pancreatic injury in AP model mice. The underlying mechanism of THBS1 involves the mediation of macrophage efferocytosis through specific binding to the downstream receptor CD36. Inhibition of THBS1 has been demonstrated to alleviate joint damage in murine models of rheumatoid arthritis (RA). Conclusions and Implications Targeting the THBS1/CD47 axis has substantial clinical implications for the treatment of acute pancreatitis (AP).
