The expression and clinical significance of autophagy- related genes Beclin1 and PTEN in adenomyosis

Abstract
Objective:To study the relationship between the expression of autophagy- related genes Beclin1 and PTEN and adenomyosis and to provide theoretical basis for the development and clinical treatment of adenomyosis.Methods The expression of autophagy- related genes Beclin1 and PTEN was detected with immunohistochemistry in 30 cases with eutopic endometrium(16 cases at hyperplasia phase,14 cases at secretory phase),30 cases with ectopic endometrium(17 cases at hyperplasia phase,13 cases at secretory phase) and 32 cases of normal endometrium as contasat group(22 cases at hyperplasia phase,10 cases at secretion phase).Results(1) The positive expression rate of Beclin1 in endometrium in experimental group was apparently lower than that in contrast group and the difference was statiscally significant(P 0.0167).The expression rate of Beclin1 in the ectopic endometrium,was significantly lower than that in ectopic and normal endometrium,it has statistically significant difference between these three groups(P 0.0167);there was no statistical significance of Beclin1 in the eutopic endometrium,ectopic endometrial secretory phase and proliferative phase expression(P 0.05).(2) The expression of PTEN in ectopic endometrium was the lowest in these three groups.The PTEN expression in normal endometrium was the highest in them and in ectopic endometrium of adenomyosis it was the lowest(P 0.0167).The PTEN expression of eutopic endometrium of adenomyosis was between that of normal endometrium group and that of ectopic endometrium of adenomyosis group(P 0.0167).The positive expression rate of PTEN in ectopic and ectopic endometrial hyperplasia period was below that of the secretory phase,and there was significant difference(P 0.05).(3) The protein expressions of Beclin1 and PTEN were positively correlated(rs = 0.633,P 0.01).Conclusion(1) In the ectopic endometrium,the negative expression of Beclin1 and PTEN is significantly lower than that in ectopic and normal endometrium,they may be associated with the development of adenomyosis.(2) The expression of Beclin1 and PTEN was positively correlated,which plays an important role in the pathogenesis and development of adenomyosis.Probably due to the loss of Beclin1 and PTEN protein,the occurrence and development of human adenomyosis is promoted.
