Development of an experimental model of endometriosis using mice that differentially express ectopic endometrial proteins

Abstract
Objective: To establish a homologous mouse model of endometriosis to analyze the differential expression of the ectopic endometrial proteins.Methods: Homogenous endometrial segments(5mm each segment) obtained from mice was transplanted onto right lateral uterus,abdominal wall and small intestinal membrane,respectively.The recipient mice were raised for 30,60 and 90 days,and then were euthanized,respectively.Endometriotic lesions that developed in the uterus,abdomen and intestine were then examined.The differential expression of the proteins in the recipient mice for 180 days was analyzed by 2D electrophoresis.Results: The transplanted ratio of the endometrium was 25.0%,50.0% and 84.2%,respectively,in the recipient mice for 30,90 and 180 days.The heterotopic endometrial lesions at different tissues were more clearly existing in clear cyst.Historically,endometriotic lesions at uterus were sharply higher than at the abdomen and intestine.The 2D electrophoresis indicated that the normal endometrium had 99 protein dots that were the same as the endometriotic lesions.The endometriotic lesions showed 13 differential dots that were as 5 times brighter as the normal endometrium in the abundance.The endometriotic lesions expressed more quantity and types of proteins than the normal endometrium.Conclusion: The present endometriosis model using mice would be useful for expeditious quantitative evaluation of extopic endometriotic proteins.
