Non-steroidal antiinflammatory\ndrugs (NSAIDs), widely used due to their\nanalgesic and anti-inflammatory properties, consistently\ninhibit ovulation in all mammalian species investigated\nso far, likely due to the inhibition of cyclooxygenase 2\n(COX-2), the inducible isoform of COX, that is the ratelimiting\nenzyme in prostaglandin (PG) synthesis. COX-2\ninhibition has major effects on ovulation, fertilization\nand implantation, and NSAID therapy is likely\nimplicated in human infertility and could be an\nimportant, frequently overlooked, cause of ovulatory\ndisfunction in women. Although there is compelling\nevidence for a role of PGs in ovulation, the molecular\ntargets and the precise role of these compounds in the\novulatory process are not fully understood.\nMorphological studies from rats treated with\nindomethacin (INDO), a potent inhibitor of PG\nsynthesis, provide evidence on the actions of NSAIDs in\novulation, as well as on the posible roles of PGs in the\novulatory process. Cycling rats treated with INDO\nduring the preovulatory period show abnormal\novulation, due to disruption of the spatial targeting of\nfollicle rupture at the apex. Noticeably, gonadotropinprimed\nimmature rats (widely used as a model for the\nstudy of ovulation) show age-dependent ovulatory\ndefects similar to those of cycling rats treated with\nINDO. These data suggest that NSAID treatment\ndisrupts physiological mechanisms underlying spatial\ntargeting of follicle rupture at the apex, which are not\nfully established in very young rats.
