This review explores the intricate molecular mechanisms underlying ferroptosis, emphasizing its reliance on iron metabolism and oxidative stress, which disrupt key processes in reproductive tissues, including granulosa cell function, folliculogenesis, and embryo implantation. Increasing evidence linking ferroptosis to these conditions offers new therapeutic opportunities, with iron chelators, lipid peroxidation inhibitors, and antioxidants showing the potential to alleviate reproductive dysfunction by modulating ferroptotic pathways. In ovarian cancer, ferroptosis inducers combined with conventional cancer therapies, such as chemotherapy, provide promising strategies to overcome drug resistance. This review synthesizes current knowledge on ferroptosis and highlights its importance as a therapeutic target in reproductive health, emphasizing the need for further research to refine and expand treatment options, evaluate their applicability in clinical settings, and explore their role in fertility preservation. By advancing our understanding of ferroptosis regulation, these therapeutic approaches could lead to novel treatments for reproductive disorders and cancers, offering new hope for improving outcomes in women’s health and cancer therapy.\nSimilar content being viewed by others\nData availability\nNot applicable.\nAbbreviations\n- PCOS:\n-\nPolycystic ovary syndrome\n- POI:\n-\nPremature ovarian insufficiency\n- GPX4:\n-\nGlutathione peroxidase 4\n- GSH:\n-\nGlutathione\n- LPCAT3:\n-\nLysophosphatidylcholine acyltransferase 3\n- NCOA4:\n-\nNuclear receptor coactivator 4\n- ACSL4:\n-\nAcyl-CoA synthetase long-chain family member 4\n- TfR1:\n-\nTransferrin receptor 1\n- FPN:\n-\nFerroportin\n- PUFAs:\n-\nPolyunsaturated fatty acids\n- ROS:\n-\nReactive oxygen species\n- ALOX15:\n-\nArachidonate 15-lipoxygenase\n- LOX:\n-\nLipoxygenase\n- LIP:\n-\nLabile iron pool\n- IREB2:\n-\nIron-responsive element-binding protein 2\n- Tf:\n-\nTransferrin\n- FTH1:\n-\nFerritin heavy chain 1\n- FSH:\n-\nFollicle-stimulating hormone\n- LH:\n-\nLuteinizing hormone\n- NCOA4:\n-\nNuclear receptor coactivator 4\n- FSP1:\n-\nFerroptosis suppressor protein 1\n- ACSL4:\n-\nAcyl-CoA synthetase long-chain family member 4\n- SCD1:\n-\nStearoyl-CoA desaturase 1\n- CD44:\n-\nCluster of differentiation 44\n- HO-1:\n-\nHeme oxygenase-1\n- NRF2:\n-\nNuclear factor erythroid 2-related factor 2\nReferences\nStockwell BR, Friedmann Angeli JP, Bayir H, Bush AI, Conrad M, Dixon SJ, Fulda S, Gascón S, Hatzios SK, Kagan VE et al (2017) Ferroptosis: a regulated cell death nexus linking metabolism, redox biology, and disease.
